I'm thinking it must have been by BB King, since all the time you're hearing people say they'd rather hear one note by BB King than "n" notes by "x" number of other guitarists. But not to leave anyone out, maybe there should be two categories.

    a) best note played by BB King. I mean he must have had some days that were better than others.
    b) best note not played by BB King, which is understood to be worse than all notes played by BB King. Of course I don't mean "notes which BB King chose not to play". I really mean "notes that people who are not BB King did choose to play".

The first specific note that comes to my mind is that super high note David Gilmour hits in the second solo of Comfortably Numb.

    Reference 6:21

The more I think about it, it might be the high note at 2:43 that John Popper hits in his harmonica solo on The Hook

The note slipped into your desk in the second grade by your cute little classmate, Mindy, saying that she wanted to kiss you.

I think John McLaughlin hit it several times, but only for a few milliseconds.

The very first note in "I Feel Fine" by The Beatles. I've never heard anyone duplicate that note.

It's actually the space between the notes that is the best .

    The opening of Purple Haze.

The pinch harmonic Joe Walsh plays in the Already gone solo.

That note in the Smoke on the water solo right as Blackmore modulates up.
