Now greatest can mean a number of things. Iconic, historic, innovative, best... You decide. But hands down we would all have to agree the greatest use of an effect in music history is....

Wah Wah -- "Theme From Shaft"

What examples you got for second place?

    Rockman Chorus?

hang on, you're not asking about the greatest use of a certain kind of effect, but the greatest effect use EVER

hard to say, but this phaser-like sound sure holds up

Some good ones listed so far.
I’d have to put Waylon’s phaser into the mix somewhere. Before him, who’d have thought you could have an effect like that in country music.

I was gonna cite this song too!

On the (longer) album version of "Crimson and Clover" the guitar solo features choruses of Tremolo, wah-wah, delay, and fuzz- all in turn.
    Love it!

I know Voodoo Chile was posted....I like this live version....That neck looks like a skinny broomstick in Hendrix' hands.

    Yes, some fantastic examples given.

I'll add the very famous "bagpipe" guitars that Big Country used -- courtesy of a MXR Pitch Transposer.

    This....

The times I saw Jeff Beck and Spirit live where Jeff and Randy would play melody lines composed of 3 part harmonies during guitar solos using an Echoplex set for a very long delay, and timing it so when the first line repeated, they'd play a second line with it, and then again, all in perfect sync. I've seen hundreds of concerts by many of the greatest rock and blues players in history, and to this day, seeing them both do that so expertly and creatively stands as the most astonishing thing I've ever seen any of my greatest guitar heroes do.

I’ll go with Van Halen’s phaser effect ftw.
