I'm refinishing a les paul that has been previously finished with wipe on poly ,It's sanded back to the wood now
So do I need to use grain filler even though the pores were previously filled ?

If you only sanded it and didn't use a stripper you should be ok without grain filler. You can apply a few coats of sealer and sand it smooth. You'll know then if the sealer isn't enough and grain filler is required.

What you've hopefully been able to do is to leave the old filler and varnish in the pores, letting that old stuff be the pore filler. You'll know as soon as you get a coat of sealer on there whether another filler application is necessary, and you should be able to apply it between coats of sealer. BTW I hate using grain fillers, probably never going to get any good at it.
