Go check out the museums mentioned above. Those are some great suggestions. Go to Martin Music on Poplar and check out those PRS guitars!
The Gibson tour is cool, just make sure you go early so you can see them working.

+1 I spent five days there and had a great time but it's a tuff city ( and I grew up in Detroit). Take a cab everywhere. We stayed at the Holiday Inn half a mile from Beale and we walked to Beale the first night. Big mistake. After that we took cabs everywhere, the cabbies on Beale would charge us 5 bucks to take us back to our motel . The Stax museum is a must as well as the Gibson Plant. Grace land was a lot of fun. Except for BB's club Beale street seemed to be a lot of Stevie Ray Wanna bees' but it was still a good time. Sun records was great .I bought a bunch of Sun Studio picks while I was there and have given them to my guitar playing friends. They've been a big hit. I went there during the MLK assassination anniversary so the Civil right museum ( Loraine Motel) was especially moving.
